The condition is that the sponsor validates the repository on a risk basis. It must also be able to evidence the audit trail, permissions, direct inspector access and archiving for at least 25 years. A specialised eTMF makes sense once several trials, countries or partners are involved. It becomes effectively necessary when completeness, contemporaneous filing and the audit trail must be shown without manual workarounds, typically ahead of an inspection or a marketing authorisation application. Regulators do not mandate a product. What changes the answer is that the configuration, validation and maintenance burden of a general-purpose repository grows with every additional trial.

How do SharePoint, Google Drive and a specialised eTMF compare for the TMF?

General-purpose platforms provide storage, versioning and activity logs. In the EU, it must also be archived for at least 25 years after the end of the trial in a complete, legible and traceable form. For electronic TMFs, EMA and FDA additionally expect validation, a protected audit trail, role-based access and direct read-only access for inspectors. No regulation prescribes a product. What is assessed is whether the system in use can demonstrably deliver these properties.

EU Regulation 536/2014: Articles 57 and 58

Under Article 57 of Regulation (EU) No 536/2014, the TMF must at all times contain the trial’s essential documents and be readily available and directly accessible to the authorities on request. Article 58 requires sponsor and investigator to archive its content for at least 25 years after the end of the trial unless other Union law requires longer. Archive media must keep the content complete and legible throughout, and any alteration must be traceable. Any transfer of ownership must be documented. The sponsor must appoint named individuals responsible for the archives, and only they may have access.

EMA guideline on the TMF (EMA/INS/GCP/856758/2018)

The EMA guideline on the content, management and archiving of the TMF applies the same basic requirements to paper and electronic TMFs. For the primary eTMF it lists specific controls:

individual user accounts, secure passwords and role-based permissions, for example for randomisation codes and unblinded data;

locking of individual documents or the whole eTMF, regular backups and periodic test restores;

an audit trail capturing date, time and user for creation, upload, deletion and change, with an explanation where necessary;

validation under formal procedures, training for all users and formally defined metadata;

an overall index, one primary TMF system and as few satellite systems as possible;

timely filing against timelines set in SOPs or TMF plans, plus risk-based QC including audit trail review;

archiving including the audit trail, logged read-only access to the archived eTMF and validated migration including metadata.

Where duties are outsourced to a CRO, the sponsor remains responsible. It needs access to the CRO-held part, for instance through remote access to the CRO’s eTMF. The contract should cover which party holds which part, how the TMF is structured and how regulators get access. It should also set out arrangements at trial completion and what happens if a party ceases trading.

EMA guideline on computerised systems (2023)

The EMA guideline on computerised systems and electronic data in clinical trials (EMA/INS/GCP/112288/2023, in effect since September 2023) explicitly lists eTMFs and document management systems within scope. The points that matter for this decision:

Systems used for purposes other than those they were developed for, or outside the supplier’s specification, are considered inherently higher risk.

Even off-the-shelf, non-trial-specific products need a risk assessment of whether they are fit for their intended use. Systems must be validated whether they are commercially available or provided as a service.

Ordinary users must not be able to deactivate audit trails. Audit trails must be protected from change and should be stored within the system. They must show what changed, by whom, when and, where applicable, why.

Inspectors must be given full, direct, read-only access with a unique identification on request. Shared accounts are unacceptable.

Cloud providers must be qualified and contracts must be detailed. Providers unwilling to support audits or inspections should not be used.

Validation of a data migration deserves no less focus than system validation, and data and audit trail should not be separated.

ICH E6(R3): essential records

ICH E6(R3) speaks of essential records and describes them in Appendix C on a risk-proportionate basis, so their nature and extent depend on trial design and conduct. The Principles and Annex 1 have applied in the EU since 23 July 2025, and Annex 2 takes effect there on 15 January 2027. Sponsors should retain essential records securely, report transfers of ownership and apply computerised system requirements proportionately. Monitoring may include direct, read-only remote access to the systems in which essential records are retained.

United States: 21 CFR Part 11, Part 312 and the 2024 FDA Q&A

21 CFR 11.10(e) requires secure, computer-generated, time-stamped audit trails that independently record operator entries and actions that create, modify or delete records. Subpart C covers electronic signatures, including the signer’s name, the date and time, the meaning of the signature and its link to the record. A TMF audit trail, by contrast, must be available for as long as the records under 21 CFR 11.10(e) and archived with the TMF under the EMA guideline. Without a validated export, evidence disappears before an inspection is even announced.

Versioning: Microsoft Learn states that the default for new SharePoint Online libraries is 500 major versions with no expiry (vendor information). EMA expects superseded versions to be kept so the trial can be reconstructed. Version limits and automatic trimming therefore need deliberate configuration and validation.

Structure and metadata: Folders are not an index. Without mandatory metadata per artefact, filing, retrieval and QC depend on individual diligence. EMA also states that file names should not replace version details.

Completeness and timeliness: General-purpose platforms have no concept of expected documents. Completeness and contemporaneous filing can only be evidenced through a separately maintained list.

Inspector access: EMA expects direct read-only access with a unique identification. A guest account limited to some folders, or a staff member steering the inspector, meets this only in part.

Partner access: Link sharing and shared accounts are quick to set up. They contradict the principle of individual accounts and make it hard to prove who saw or changed what, and when.

Archive and exit: Over 25 years, licence models, platforms and perhaps the company itself will change. Without a format strategy, a named archivist and exit arrangements, the Article 58 legibility duty is hard to evidence.

GMP document control, meaning SOPs, approvals and training records, has different priorities. That trade-off is covered on the SharePoint vs. eDMS page. Functional criteria are set out under eDMS requirements.

What do inspection findings show about TMFs and systems?

Essential documents are among the most frequently cited areas in recent EU inspections. EMA’s 2024 report explicitly gives as an example the lack of access for monitors and inspectors to trial-relevant electronic systems containing essential documents. Inspection reports do not name products, however, so public data cannot show that findings stem specifically from SharePoint or Google Drive. What they do show is that the properties criticised are exactly those a general-purpose repository lacks without configuration.

EMA, 2024 annual report: The GCP Inspectors Working Group report for 2024 (EMA/INS/GCP/37608/2025) was adopted by the GCP IWG on 15 December 2025 and published in January 2026. It covers 67 CHMP-requested inspections: 52 routine and 15 for-cause, of which 18 were at sponsors, 42 at investigator sites and 5 at CROs. It records 335 critical and major findings (11 critical, 324 major), and minor findings are no longer included. The essential documents sub-category accounts for 41 findings (39 major, 2 critical). The computer system category accounts for 14, of which 12 were at sponsors. Examples given include:

lack of access to electronic systems containing essential documents;

incomplete TMFs;

no formally documented TMF QC process;

no established process for certifying copies;

inadequate data migration and system validation.

CHMP-requested inspections are only a small fraction of all EU inspections.

MHRA: The MHRA Inspectorate Blog (2015) describes the TMF as the story of how a trial was conducted. It questions the common practice of expert users steering inspectors through the eTMF. An MHRA presentation from 2016 reports TMF findings at 84% of 22 sponsor inspections (2013–2014) and 82% of 11 sponsor inspections (2014–2015). It also cites the definition of a critical finding: a TMF that is not readily available, not accessible or so incomplete that it cannot form the basis of inspection. No more recent TMF-specific MHRA percentages could be found.

Industry surveys (vendor data): In the Veeva Paperless TMF Survey 2016, the share of sponsors using a local file system as their eTMF fell from 26% (2014) to 8% (2016). The Veeva Unified Clinical Operations Survey 2020 found 68% of responding sponsors and CROs using a purpose-built eTMF application, against 17% in 2014. These are vendor surveys with pre-2023 data that lean towards larger organisations.

Practical steps for staying inspection-ready are covered in the TMF inspection readiness guide.

When is SharePoint or Google Drive enough for the TMF?

A general-purpose repository can be defensible when the trial is small and contained, or when the TMF sits entirely in a full-service CRO’s validated eTMF. Neither EMA nor FDA rules out general IT or cloud services. Both require a documented risk assessment, validation proportionate to risk and contracts that secure access for the full retention period. A sponsor that maintains this evidence properly can start with SharePoint or Drive.

Situations where general storage can be defensible

A single early-phase trial, such as first-in-human, with one or a few sites and low document volume.

A full-service CRO holds the complete TMF in its own eTMF, and the sponsor keeps only sponsor-specific records or working copies.

A small, experienced team that files and checks continuously and does not need Part 11 signatures inside the repository.

The minimum you then need

Risk assessment and validation: document TMF requirements and assess risks. Test critical functions such as permissions, versioning, audit logging and export, and assess platform updates under change control. The 2024 FDA Q&A allows validation scaled to the nature of the system. EMA holds the user accountable even for commercial products.

SOP, TMF plan and index: structure based on the TMF Reference Model, filing timelines, naming and metadata conventions, and responsibilities.

Access concept: individual accounts, least privilege, segregated areas for unblinded data and documented periodic reviews.

Audit trail evidence: check the licence tier and log retention. Export and archive audit logs through a validated process before the vendor’s retention period expires.

Versioning and backup: configure version limits deliberately, keep separate backups and document restore tests.

Inspector access: a prepared read-only role with an individual account, tested before inspection.

Archiving and exit plan: a named archivist, a format strategy and an export routine with metadata. Add provisions for changing provider or for the company ceasing to exist.

Supplier qualification: assessment of the cloud provider and contracts covering data location, access, incidents and data return, as described in Annex 1 of the EMA guideline and the 2024 FDA Q&A.

What the sponsor must still ensure when the CRO holds the TMF

Even when the CRO maintains the TMF, the sponsor remains responsible for it under the EMA guideline and ICH E6(R3). It needs regular or remote access to the CRO-held part and contractual terms on structure, regulator access and handover at the end of the trial. It also needs oversight, for example through audits, and archiving for the full period. If the TMF is handed over at the end, the transfer must be validated and must preserve metadata and audit trail.

What makes an eTMF simple to use?

A simple eTMF comes with the TMF Reference Model preconfigured, enforces metadata on upload and measures completeness automatically. It also offers roles for sites, CROs and inspectors, so the sponsor does not have to build and validate these functions itself. Simple does not mean feature-poor. It means set-up, day-to-day use and validation evidence remain manageable for a small team. There are no robust independent figures on typical set-up time, so vendor claims should be evidenced during selection.

Preconfigured Reference Model: The TMF Reference Model is the de facto standard for TMF structure and is now stewarded by CDISC. The current published release is version 3.3.1 of 11 August 2023. A 2024 CDISC presentation describes it as 11 zones, 48 sections and 249 artefacts. Its successor will not be issued as version 4 but as the CDISC TMF Standard Model v1, which is still in community review in November 2026. The project timeline reported by Just in Time GCP from the CDISC Europe Interchange in May 2026 plans its release for April 2027.

Vendor validation package: Under the risk-based approach of GAMP 5 Second Edition (ISPE, 2022), sponsors can leverage supplier evidence such as IQ/OQ documentation and test reports. The EMA guideline expects the sponsor to assess this evidence, typically through an audit, and to close gaps with its own testing.

Usability for partners: guest access and upload functions for sites and CROs, and where relevant an eISF link. Sites must not lose control of their own records.

Completeness and QC: dashboards for expected versus filed documents, timeliness metrics and QC workflows with status tracking.

Inspector access: a predefined read-only role with logged access.

Export and archive: export with metadata and audit trail, ideally supporting the eTMF Exchange Mechanism Standard, currently version 1.0.2 (effective January 2022).

Licensing models mainly differ in whether they charge per trial, per user or by document volume. Many add a platform fee and one-off costs for onboarding, validation support and migration. Price ranges and a total cost of ownership view are in the eTMF cost guide. A detailed vendor comparison is available in the eTMF software comparison.

How do you migrate a TMF from SharePoint to an eTMF?

A TMF migration is a process that must be validated. Documents are inventoried, mapped to the Reference Model index and enriched with metadata. They are then verified with checksums and sampling and documented in a migration report. EMA expects migration validation to receive no less focus than system validation, and data and audit trail must not be separated. With SharePoint or Drive, the most critical point is the audit log history, which expires after the vendor’s retention period.

Sensible moments to migrate

before trial start, while few documents exist;

before adding further trials, countries or CROs;

well ahead of a first inspection or marketing authorisation application, not during last-minute preparation;

when changing CRO, or when the CRO hands the TMF back to the sponsor at the end of the trial.

Approach

Inventory: capture every storage location, including email folders and drives, and identify duplicates and drafts.

Mapping: assign each file to a Reference Model artefact and retain the mapping as part of the migration record, as EMA expects.

Metadata enrichment: add document date, country, site, version and status. System timestamps are no substitute for the business document date.

Versions and audit history: bring superseded versions across. Export audit logs and version histories before the vendor’s retention period expires and archive them linked to the documents.

Migration plan and validation: analyse risks and rehearse the procedure with test data. After migration, run count reconciliation, checksum or hash comparison and risk-based sampling.

Migration report: document deviations, corrections and approval.

Source system: set it to read-only and decommission it only after verified migration. Originals may only be destroyed where certified copies exist. EMA and FDA define a certified copy as one verified by dated signature or a validated process to contain the same information as the original, including context and metadata.

Typical risks are loss of metadata and audit history, mis-filing against artefacts, duplicates, incomplete transfer and overwritten date and time stamps. The TMF often refers to SOPs, training records, deviations or CAPAs held in other systems. EMA names those systems explicitly as part of the TMF and expects the same controls for them. An integrated, pre-validated platform covering eTMF and quality processes then reduces the number of interfaces to validate. The eQMS software comparison gives an overview of quality systems.

Which option fits which starting point?

The right choice depends less on company size than on who holds the TMF, how many trials and partners are involved and when an inspection is likely. The recommendations below follow from the regulatory requirements, the vendor-documented limits of general-purpose storage and published inspection findings.

TMF held entirely by a full-service CRO

A sponsor eTMF is not mandatory if the CRO keeps the complete TMF in its validated eTMF. The contract must, however, secure remote access, regulator access, oversight and end-of-trial handover with metadata and audit trail. The sponsor must also choose a target system for 25-year archiving before handover is due. Sponsor-specific records belong in a controlled, validated repository.

Sponsor maintains the TMF, one trial

General storage is defensible here if the minimum package is in place: validation, SOP, TMF plan, access concept, audit log export, inspector role and archive plan. Once configuration, validation and upkeep are counted honestly, a pre-validated eTMF is often the simpler route even for one trial. This applies particularly where Part 11 signatures or US submissions are planned.

Several trials or countries

With several trials, countries or CROs, a specialised eTMF is usually the more robust option. In general storage, completeness tracking, consistent metadata, partner access and QC need more and more manual effort, and error rates rise. These are precisely the areas that appear in inspection findings.

Inspection or marketing authorisation application ahead

Ahead of an inspection or application, the TMF must be complete and contemporaneous, and inspectors must be able to read it directly without a guide. Teams still on SharePoint or Drive should complete a validated migration with enough lead time. If that is not feasible, harden the current repository: secure the audit logs, reconcile the index against the expected-document list and test inspector access.

Frequently asked questions about SharePoint, Google Drive and eTMF

Can SharePoint be used as an eTMF?

Yes, if SharePoint is validated on a risk basis for TMF use and configured so that index, metadata, permissions, audit trail, inspector access and archiving meet EMA and FDA expectations. According to Microsoft Learn, Purview Audit (Standard) keeps audit records for 180 days (for logs generated on or after 17 October 2023). A higher licence tier or a validated log export is therefore needed. The effort rests entirely with the sponsor and grows with every trial.

Is Google Drive GCP compliant?

No system is GCP compliant on its own. Compliance comes from validated use within suitable processes. Google Drive can be part of such a set-up, but Google’s Admin Help page “Data retention and lag times” lists 6 months’ retention for Drive log events. TMF structure, mandatory metadata and completeness tracking are also missing. Without additional controls, validation and log archiving, Drive does not meet TMF expectations.

What simple software is suitable for clinical trial documents?

For a TMF, choose software that ships with the TMF Reference Model preconfigured and enforces metadata on upload. It should also measure completeness, keep a durable audit trail and offer read-only inspector access. A vendor validation package reduces your own effort but does not replace the sponsor’s assessment. General-purpose storage suits working documents, but not the TMF without further controls.

Do I need an eTMF if my CRO maintains the TMF?

Not necessarily, if the CRO keeps the complete TMF in its validated eTMF. The sponsor remains responsible, though. It needs access, oversight and a contractual end-of-trial handover with metadata and audit trail. In the EU it must also ensure archiving for at least 25 years. By handover at the latest, it needs a suitable target system.

What is the difference between an eTMF and a CTMS?

An eTMF manages the essential records that show how a trial was conducted and archives them in a form inspectors can review. A CTMS runs day-to-day trial operations such as milestones, sites, recruitment and monitoring visits. EMA notes that reports generated from CTMS data may be filed in the eTMF as PDFs. The original dynamic file then stays in the source system.

How do I migrate a TMF from SharePoint to an eTMF?

Inventory the content, map it to the Reference Model index and enrich the metadata. Export audit logs before the vendor’s retention period expires and validate the migration with test data. Afterwards, verify it with count reconciliation, checksums and sampling, and record the outcome in a migration report. Decommission the source only after verified migration, and destroy originals only where certified copies exist.

How long must the TMF be retained?

In the EU, at least 25 years after the end of the trial under Article 58 of Regulation (EU) No 536/2014, unless other Union law requires longer. In the US, 21 CFR 312.57(c) requires sponsors to keep records for two years after approval or after investigational supply is discontinued. For multi-region programmes, the longest applicable period governs in practice.

Does the eTMF vendor have to take over validation?

No. Vendors typically provide validation documentation. The sponsor must assess it under the EMA guideline and supplement it with its own testing where gaps exist. Responsibility for validating the system in use stays with the sponsor, including for cloud and SaaS solutions.

Sources

<a
